We report on the development of a fiber-based laser transmitter designed for active remote sensing spectroscopy. The transmitter uses a master oscillator power amplifier (MOPA) configuration with a distributed feedback diode-laser master oscillator and an erbium-doped fiber amplifier. The output from the MOPA is frequency-doubled with a periodically poled potassium titanium oxide phosphate crystal. With 35 W of single-frequency peak optical pump power, 8 W of frequency-doubled peak power was achieved. The utility of this single-frequency, wavelength tunable, power scalable laser was then demonstrated in a spectroscopic measurement of diatomic oxygen A band. 相似文献

Laser Doppler imaging has been widely used for the evaluation of cutaneous blood flow. We report on how the self-mixing interferometry configuration with a laser diode is explored for what is believed to be the first time to generate flow maps. The experiment was carried out by sensing the laser intensity power spectrum at each pixel as the laser was scanned over a model that mimics the properties of skin and circulating blood. 相似文献

We report the design, synthesis, characterization, and operation of a [2]rotaxane in which a palladium-complexed macrocycle can be translocated between 4-dimethylaminopyridine and pyridine monodentate ligand sites via reversible protonation, the metal remaining coordinated to the macrocycle throughout. The substitution pattern of the ligands and the kinetic stability of the Pd-N bond means that changing the chemical state of the thread does not automatically cause a change in the macrocycle's position in the absence of an additional input (heat and/or coordinating solvent/anion). Accordingly, under ambient conditions any of the four sets of protonated and neutral, stable, and metastable co-conformers of the [2]rotaxane can be selected, manipulated, isolated, and characterized. 相似文献

The ability of multiple spatial pathways to sum coherently and facilitate charge transfer is examined theoretically. The role of multiple spatial pathways in mediating charge transfer has been invoked several times in the recent literature while discussing charge transfer in proteins, while multiple spatial pathways are known to contribute to charge transport in metal-molecule-metal junctions. We look at scaling laws for charge transfer in donor-bridge-acceptor (D-B-A) molecules and show that these scaling laws change significantly when environment-induced dephasing is included. In some cases, D-B-A systems are expected to show no enhancement in the rate of charge transfer with the addition of multiple degenerate pathways. The origins of these different scaling laws are investigated by looking at which Liouville space pathways are active in different dephasing regimes. 相似文献

A sensitive and specific assay aimed at measuring the oxidized nucleic acids, 8-oxoguanine (8-oxoGua), fapy-guanine (Fapy-Gua), 8-oxoguanosine (8-oxoGuo), 8-oxo-2'-deoxyguanosine (8-oxodG) has been developed by coupling reversed phase liquid chromatography (HPLC) with electrospray tandem mass spectrometry detection (MS/MS) and isotope dilution. The HPLC-MS/MS approach with multiple reaction monitoring (MRM) allowed for the sensitive determination of 8-oxoGua, Fapy-Gua, 8-oxoGuo, and 8-oxodG in human urine samples. There is no sample preparation needed except for the addition of buffer and (13)C- and (15)N-labeled internal standards to the urine prior to sample injection into the HPLC-MS/MS system. This method was tested in urine samples from non-smokers, smokers, non-smokers with chronic kidney disease (CKD) and smokers with CKD, to assess the level of oxidative damage to nucleic acids. Markers of both RNA and DNA damage were significantly increased in the smokers with and without CKD compared to their respective control subjects. These findings suggest that a highly specific and sensitive analytical method such as isotope dilution HPLC-MS/MS may represent a valuable tool for the measurement of oxidative stress in human subjects. 相似文献

Friction force measurements have been conducted with a colloid probe on mica and silica (both hydrophilic and hydrophobized) after long (24 h) exposure to high-humidity air. Adhesion and friction measurements have also been performed on cellulose substrates. The long exposure to high humidity led to a large hysteresis between loading and unloading in the friction measurements with separation occurring at large negative applied loads. The large hysteresis in the friction-load relationship is attributed to a contact area hysteresis of the capillary condensate which built up during loading and did not evaporate during the unloading regime. The magnitude of the friction force varied dramatically between substrates and was lowest on the mica substrate and highest on the hydrophilic silica substrate, with the hydrophobized silica and cellulose being intermediate. The adhesion due to capillary forces on cellulose was small compared to that on the other substrates, due to the greater roughness of these surfaces. 相似文献

The conditions for reverse-micelle formation were studied for solutions of tetra-n-octyldiglycolamide (TODGA) in alkane diluents equilibrated with aqueous solutions of nitric or hydrochloric acids in the presence and absence of Nd3+. Small-angle neutron scattering, vapor-pressure osmometry, and tensiometry are all consistent with the partial formation of TODGA dimers at the lowest acidities, transitioning to a polydisperse mixture containing TODGA monomers, dimers, and small reverse-micelles of TODGA tetramers at aqueous nitric acid acidities of 0.7 M or higher in the absence of Nd. Application of the Baxter model to the samples containing 0.005-0.015 M Nd reveals the persistence of tetrameric TODGA reverse-micelles with significant interparticle attraction between the polar cores of the micelles that increases with increasing organic phase concentrations of acid or Nd. Our experimental findings suggest that the peculiar behavior of TODGA with respect to the extraction of trivalent lanthanide and actinide cations arises from the affinity of these metal cations for the preformed TODGA reverse-micelle tetramers. 相似文献

Quantitative electron paramagnetic resonance (EPR) method was applied to characterise radicals stabilised in polyphenolic matrices of various biogenic materials: lichens, mosses, composts, soils, peats, brown coals and sewage sludge sediments. The investigations were carried out on raw materials and extracted fractions of humic acids (HA). General trends of g value and spin concentration changes were found. These parameters in lichens strongly depend on lichen species and air pollution. Determination of the g value and semiquinone spin concentration allows to assess degree of transformation of organic matter in compost, soil, peat and lignite. Application of gaseous ammonia as a base penetrating the organic matrices extends the possibilities and usefulness of the method. Interaction of metal ions with humic materials is illustrated by interaction of VO2+ ion with peat and lignite HA as well as demineralised (raw and carbonised) brown coal. Our investigations demonstrate that quantitative EPR is a rapid and effective monitoring method to study the influence of various environmental factors on substances containing polyphenolic matrices. 相似文献

The structure of N-linked glycans is determined by a complex, anabolic, intracellular pathway but the exact role of individual glycans is not always clear. Characterization of carbohydrates attached to glycoproteins is essential to aid understanding of this complex area of biology. Specific mass spectral detection of glycopeptides from protein digests may be achieved by on-line HPLC-MS, with selected ion monitoring (SIM) for diagnostic product ions generated by cone voltage fragmentation, or by precursor ion scanning for terminal saccharide product ions, which can yield the same information more rapidly. When glycosylation is heterogeneous, however, these approaches can result in spectra that are complex and poorly resolved. We have developed methodology, based around precursor ion scanning for ions of high m/z, that allows site specific detection and structural characterization of glycans at high sensitivity and resolution. These methods have been developed using the standard glycoprotein, fetuin, and subsequently applied to the analysis of the N-linked glycans attached to the scrapie-associated prion protein, PrP(Sc). These glycans are highly heterogeneous and over 30 structures have been identified and characterized site specifically. Product ion spectra have been obtained on many glycopeptides confirming structure assignments. The glycans are highly fucosylated and carry Lewis X or sialyl Lewis X epitopes and the structures are in-line with previous results. 相似文献

Considerable attention is typically given to radiation safety in the design of irradiators and initially establishing the program. However, one component that may not receive enough attention is applying the continuous improvement philosophy to the radiation safety program. Periodic total program assessments of radiation safety can ensure that the design and implementation of the program continues to be applicable to the operations. The first step in the process must be to determine what is to be covered in the program assessment. While regulatory compliance audits are a component, the most useful evaluation will extend beyond looking only at compliance and determine whether the radiation safety program is the most appropriate for the particular operation. Several aspects of the irradiator operation, not all of which may routinely be considered “radiation safety”, per se, should be included: Design aspects of the irradiator and operating system, system controls, and maintenance procedures, as well as the more traditional radiation safety program components such as surveys, measurements and training. 相似文献
